Bremelanotide is a cyclic heptapeptide that binds several melanocortin receptors rather than one. In vitro assays report agonist activity at MC1R, MC3R, MC4R and MC5R, with MC4R generally treated as the subtype most relevant to sexual effects. MC4R is expressed in hypothalamic nuclei involved in appetite, energy balance and motivated behaviour, which provides a plausible route for central action. Selective MC4R agonists studied in animals produce comparable behavioural changes, supporting that interpretation.
How receptor activation translates into a change in desire is not established in detail. Proposed steps include modulation of dopaminergic signalling in reward circuits and downstream effects on autonomic tone. Human data consist mainly of clinical trials measuring self-reported outcomes rather than direct measurements of brain activity or transmitter release. The transient rise in blood pressure sometimes observed after administration is likewise reported consistently but explained only partly by known melanocortin pathways.
Melanocortin receptors form a family of five G-protein-coupled proteins, labelled MC1R through MC5R, that respond to peptides derived from pro-opiomelanocortin. Alpha-melanocyte-stimulating hormone and adrenocorticotropic hormone are the best-known endogenous ligands. The receptors are distributed differently across tissues, so a single agonist can produce effects in the brain, skin, adrenal gland and vasculature. This distribution explains why one peptide can influence both pigmentation and motivated behaviour.
Regulatory status varies by jurisdiction, where approved prescription products, compounded preparations and research-grade material are treated as distinct categories with different documentation requirements. Suppliers of research material commonly issue a certificate of analysis listing purity, identity and sometimes endotoxin content. Independent verification by a third-party laboratory is often recommended because self-reported figures are difficult to check. Literature discussions usually state the source, purity and storage conditions of the material used, since these details affect reproducibility. Analysts note that a reported purity figure does not by itself describe biological activity.
Lyophilised peptide is generally held below minus twenty degrees Celsius, protected from light and moisture, because hydrolysis and oxidation accumulate faster at ambient temperature. Once reconstituted, solutions are typically kept between two and eight degrees Celsius and used within a short window defined by the supplier. Repeated freeze-thaw cycles are avoided since they promote aggregation and loss of soluble material. Container material matters as well, because peptides adsorb to certain plastics and glass surfaces at low concentration. Stability figures supplied by a vendor apply only to the specific lot and buffer that were tested.

Whether the behavioral effect originates centrally, peripherally, or through both remains an active question. Animal experiments using receptor antagonists and site-specific injections point toward hypothalamic melanocortin circuits as a key locus, but translating those findings to humans is not straightforward. Blood pressure changes observed in trials suggest a vascular component that may be peripherally mediated. The relationship between receptor occupancy and reported effect has not been mapped in humans, and no validated biomarker predicts response. This gap makes it difficult to explain individual variability on pharmacological grounds alone.
Bremelanotide acts as an agonist at melanocortin receptors, a family of five G-protein-coupled receptors labeled MC1 through MC5. Binding studies indicate activity at several of these subtypes rather than strict selectivity for one. Signalling proceeds mainly through Gs-mediated activation of adenylyl cyclase, raising intracellular cyclic AMP. The MC4 receptor, expressed in hypothalamic and limbic circuits, is widely regarded as the subtype most relevant to sexual response. Because the molecule is not subtype-selective, effects at other melanocortin receptors are expected and are used to explain some observed side effects.
Bremelanotide is a moderately large peptide with a molecular mass near 1025 daltons. In lyophilised form it appears as a white to off-white powder and is freely soluble in water and other polar solvents. The intact lactam ring is essential for receptor affinity, while linearised fragments bind far more weakly. Solutions are sensitive to extremes of pH and to prolonged exposure to light and heat, so handling typically involves buffered conditions and cold storage. Its short plasma half-life reflects rapid distribution and clearance rather than chemical breakdown inside the vial.
Development of the peptide passed through several delivery formats, including an intranasal version tested in early trials and an injectable version that entered later clinical study. Regulatory approval for a subcutaneous product in the United States was granted in 2019 after review of controlled trials in premenopausal women. Outside the clinic, the compound circulates in research and non-pharmaceutical markets under its code name, where identity and purity vary considerably between suppliers. Synonyms appearing across technical literature include bremelanotide, PT-141, and Palatin 141.
PT-141 is the research designation for bremelanotide, a synthetic cyclic heptapeptide developed as a melanocortin receptor agonist. The peptide contains seven amino acids with an internal lactam bridge that constrains the backbone into a stable ring. Cyclisation of this kind improves resistance to enzymatic degradation relative to linear analogues. The compound was first explored for effects on melanin production, because several melanocortin receptors influence pigmentation. Its later association with sexual desire emerged from observations made during that early work, and the molecule became the subject of a separate development programme.
Bremelanotide is a synthetic cyclic heptapeptide that acts on a family of G-protein-coupled receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide associated with pigmentation and several central signalling pathways. A lactam bridge constrains the ring and slows enzymatic breakdown, which distinguishes it from the linear parent molecule. Research interest moved over time from pigment biology toward central nervous system effects, particularly circuits connected to sexual desire. Parenteral delivery is used because oral bioavailability is poor.
Early clinical work used an intranasal formulation, and later programmes switched to subcutaneous delivery for more consistent absorption. A subcutaneous product received regulatory approval in the United States in 2019 for premenopausal women with acquired, generalised hypoactive sexual desire disorder. Approval followed phase 3 trials in which active treatment separated from placebo on desire and distress measures, though the average difference was modest. Labeling carries a caution about transient blood pressure elevation, so cardiovascular history is assessed before prescribing. Questions about durability of benefit beyond several months remain open.
Reverse-phase high-performance liquid chromatography is the standard method for assessing purity. Mass spectrometry confirms molecular identity and detects sequence variants or truncation products. Ultraviolet absorbance at 214 or 280 nm is used for quantification, with the choice depending on the peptide sequence. Method validation typically addresses linearity, limit of detection, and precision across a defined concentration range. Impurity profiling may also employ ion-exchange or size-exclusion chromatography, and these techniques complement one another.
Certificate of analysis documents usually report purity percentage, molecular weight, and appearance. Researchers verify identity by comparing observed and theoretical mass values. Chain-of-custody records and batch numbers help trace material from source to experiment. Independent verification of supplier claims is considered good practice when material identity is critical to a study. Records should include the storage history of each aliquot, and unexpected deviations in appearance or solubility warrant re-testing before use.
Lyophilized peptide material is generally stored at -20 °C or below to limit degradation, while reconstituted solutions are less stable and are typically kept refrigerated and protected from light. Repeated freeze-thaw cycles can accelerate aggregation and should be minimized. Stability for any specific lot depends on purity, moisture content, and packaging. Handling in a temperature-controlled environment reduces variability across replicates, and exposure to ambient humidity during weighing can introduce error. Aliquotting reduces the number of times a stock container is opened.
=== 1987 === 19 June: Heather West (16). Heather was likely to have been murdered because the Wests considered her efforts to leave the household as a threat, since she had divulged to her classmates the extensive physical and sexual abuse that occurred at Cromwell Street. Fred claimed he had not intended to kill his daughter, but carpet fibres found on two lengths of rope discovered with her remains suggested that she had been restrained and subjected to a sexual assault prior to her murder. Her body was dismembered with a heavy serrated knife and later buried in a hole in the garden, which Fred got his son to dig under the pretence of installing a fish pond. The 1994 police investigation into Heather's disappearance led to the discovery of her body and the arrest of both her parents.
==== Cholesterol synthesis ==== The cytosolic acetyl-CoA can also condense with acetoacetyl-CoA to form 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) which is the rate-limiting step controlling the synthesis of cholesterol. Cholesterol can be used as is, as a structural component of cellular membranes, or it can be used to synthesize steroid hormones, bile salts, and vitamin D.
=== October === October 1 The US imposes new sanctions on Iran, targeting rail, automotive, manufacturing, and steel sectors, as well as overseas suppliers, saying that the goal is to economically isolate the Iranian government. The US military starts sending about 9,000 soldiers to the Middle East on naval ships, such as the USS Makin Island amphibious readiness group and the USS Theodore Roosevelt carrier strike group. October 2 The US imposes sanctions on Hamas' financing network, which includes two French-based funds. Alabama state representative Brett Easterbrook is arrested on charges of first-degree human trafficking and prostitution as part of a sting. At least a dozen other men are arrested. October 3 – Trump establishes a task force to examine the risks and opportunities of AI and recommend what role the federal government should play in overseeing the technology without compromising competitiveness with China. October 4 The State Department announces that the US "kicked out" two members of the Iranian delegation who attended the recent United Nations General Assembly after they ignored an order to leave the country long after the General Assembly had ended. Trump withdraws all US bombers from RAF Fairford, a British air base targeted by an attack that the UK linked to the Iranian government, due to urgent concerns about a potentially complex drone attack ordered by the IRGC as part of a plot to target the aircraft and kill American soldiers at RAF Fairford, which is loosely modeled after Ukraine's Operation Spiderweb.
Uranium-238 (238U or U-238) is the most common isotope of uranium found in nature, with a relative abundance above 99%. Unlike uranium-235, it is non-fissile, which means it cannot sustain a chain reaction in a thermal-neutron reactor. However, it is fissionable by fast neutrons, and is fertile, meaning it can be transmuted to fissile plutonium-239. 238U cannot support a chain reaction because inelastic scattering reduces neutron energy below the range where fast fission of one or more next-generation nuclei is probable. Doppler broadening of 238U's neutron absorption resonances, increasing absorption as fuel temperature increases, is also an essential negative feedback mechanism for reactor control. The isotope has a half-life of 4.463 billion years (1.408×1017 s). Due to its abundance and half-life relative rate of decay to other radioactive elements, 238U is responsible for about 40% of the radioactive heat produced within the Earth. The 238U decay chain contributes six electron anti-neutrinos per 238U nucleus (one per beta decay), resulting in a large detectable geoneutrino signal when decays occur within the Earth. The decay of 238U to daughter isotopes is extensively used in radiometric dating, particularly for material older than approximately 1 million years. Depleted uranium has an even higher concentration of the 238U isotope, and even low-enriched uranium (LEU), while having a higher proportion of the uranium-235 isotope (in comparison to depleted uranium), is still mostly 238U.
The stage was also an important part of Thomas's life from 1929 to 1934, as an actor, writer, producer and set painter. He took part in productions at Swansea Grammar School, and with the YMCA Junior Players and the Little Theatre, which was based in the Mumbles. It was also a touring company that took part in drama competitions and festivals around South Wales. Between October 1933 and March 1934, for example, Thomas and his fellow actors took part in five productions at the Mumbles theatre, as well as nine touring performances. Thomas continued with acting and production throughout his life, including his time in Laugharne, South Leigh and London (in the theatre and on radio), as well as taking part in nine stage readings of Under Milk Wood. The Shakespearian actor, John Laurie, who had worked with Thomas on both the stage and radio thought that Thomas would "have loved to have been an actor" and, had he chosen to do so, would have been "Our first real poet-dramatist since Shakespeare." Painting the sets at the Little Theatre was just one aspect of the young Thomas's interest in art. His own drawings and paintings hung in his bedroom in Cwmdonkin Drive, and his early letters reveal a broader interest in art and art theory. Thomas saw writing a poem as an act of construction "as a sculptor works at stone," later advising a student "to treat words as a craftsman does his wood or stone...hew, carve, mould, coil, polish and plane them..." Throughout his life, his friends included artists, both in Swansea and in London, as well as in America.

=== Transistors === Both chemically controlled and voltage controlled graphene transistors have been built. Graphene-based transistors could be much thinner than modern silicon devices, allowing faster and smaller configurations. Graphene exhibits a pronounced response to perpendicular external electric fields, potentially forming field-effect transistors (FET), but the absence of a band gap fundamentally limits its on-off conductance ratio to less than ~30 at room temperature. A 2006 paper proposed an all-graphene planar FET with side gates. Their devices showed changes of 2% at cryogenic temperatures. The first top-gated FET (on–off ratio of <2) was demonstrated in 2007. Graphene nanoribbons may prove generally capable of replacing silicon as a semiconductor. A patent for graphene-based electronics was issued in 2006. In 2008, researchers at MIT Lincoln Lab produced hundreds of transistors on a single chip and in 2009, very high frequency transistors were produced at Hughes Research Laboratories. A 2008 paper demonstrated a switching effect based on reversible chemical modification of the graphene layer that gives an on–off ratio of greater than six orders of magnitude. These reversible switches could potentially be employed in nonvolatile memories. IBM announced in December 2008 graphene transistors operating at GHz frequencies. In 2009, researchers demonstrated four different types of logic gates, each composed of a single graphene transistor. In May 2009, an n-type transistor complemented the prior p-type graphene transistors.
=== Hegel === Adorno's adoption of Hegelian philosophy can be traced back to his inaugural lecture in 1931, in which he postulated, "only dialectically does philosophical interpretation seem possible to me" (Gesammelte Schriften 1: 338). Hegel rejected the idea of separating methods and content, because thinking is always thinking of something; dialectics for him is "the comprehended movement of the object itself." Like Gerhard Schweppenhäuser, Adorno adopted this claim as his own and based his thinking on one of the Hegelian basic categories, determinate negation, according to which something is not abstractly negated and dissolved into zero but is preserved in a new, richer concept through its opposite. Adorno understood his Three Studies of Hegel as "preparation of a changed definition of dialectics" and that they stop "where the start should be" (Gesammelte Schriften 5: 249 f.). Adorno dedicated himself to this task in one of his later major works, Negative Dialectics (1966). The title expresses "tradition and rebellion in equal measure." Drawing from Hegelian reason's speculative dialectic, Adorno developed his own "negative" dialectic of the "non-identical".

== History == Antimicrobial use has been common practice for at least 2000 years. Ancient Egyptians and ancient Greeks used specific molds and plant extracts to treat infection. In the 19th century, microbiologists such as Louis Pasteur and Jules Francois Joubert observed antagonism between some bacteria and discussed the merits of controlling these interactions in medicine. Louis Pasteur's work in fermentation and spontaneous generation led to the distinction between anaerobic and aerobic bacteria. The information garnered by Pasteur led Joseph Lister to incorporate antiseptic methods, such as sterilizing surgical tools and debriding wounds into surgical procedures. The implementation of these antiseptic techniques drastically reduced the number of infections and subsequent deaths associated with surgical procedures. Louis Pasteur's work in microbiology also led to the development of many vaccines for life-threatening diseases such as anthrax and rabies. On September 3, 1928, Alexander Fleming returned from a vacation and discovered that a Petri dish filled with Staphylococcus was separated into colonies due to the antimicrobial fungus Penicillium rubens. Fleming and his associates struggled to isolate the antimicrobial but referenced its therapeutic potential in 1929 in the British Journal of Experimental Pathology. In 1942, Howard Florey, Ernst Chain, and Edward Abraham used Fleming's work to purify and extract penicillin for medicinal uses earning them the 1945 Nobel Prize in Medicine.
While rare, mostly abortive, parthenogenesis is not unknown in birds and eggs can be diploid, automictic and results in male offspring. Birds are solely gonochoric, meaning they have two sexes: either female or male. The sex of birds follows the Z and W sex-determination system, where the ovum determines the sex of the offspring. Male birds have two Z chromosomes (ZZ), and female birds have a W chromosome and a Z chromosome (WZ). In mammals, by contrast, the sperm determines the sex of the offspring in the X and Y sex-determination system. A complex system of disassortative mating with two morphs is involved in the white-throated sparrow Zonotrichia albicollis, where white- and tan-browed morphs of opposite sex pair, making it appear as if four sexes were involved since any individual is compatible with only a fourth of the population. In all species of birds, an individual's sex is determined at fertilisation. One 2007 study claimed to demonstrate temperature-dependent sex determination among the Australian brushturkey, for which higher temperatures during incubation resulted in a higher female-to-male sex ratio. This, however, was later proven to not be the case. These birds do not exhibit temperature-dependent sex determination, but temperature-dependent sex mortality.

The diversity of ceramide structures undoubtedly plays an important role in the unique attributes of the stratum corneum across different body sites. For example, the stratum corneum of the face is thin and flexible to accommodate different facial expressions. In contrast, the stratum corneum covering the heel of the foot is thick and rigid to protect against trauma. Matching these structural changes, there are body-site specific alterations in the epidermal lipidome, including changes in the relative abundance of the different epidermal ceramide structures. Similar to body site-specific alterations in ceramide abundance, there are also well-characterized changes in epidermal ceramide expression in patients with inflammatory skin diseases. In the hyperplastic disorder psoriasis, investigators have reported an increase in AS and NS ceramides and a decrease in EOS, AP, and NP ceramides, which may contribute to a defect in the skin's water impermeability barrier. Studying ceramide expression in atopic dermatitis and psoriasis patients, other investigators have reported that rather than focusing on ceramide classes, ceramide sphingoid base length and fatty acid chain length have the strongest influence on the likelihood of a particular ceramide structure being upregulated or downregulated in inflamed skin. Ceramide levels in the skin, hair, and nails can be reduced due to environmental changes (such as dry/polluted air), use of harsh sulfates, excessive heat (including heat styling), UV exposure, and biological aging.

Bremelanotide binds several melanocortin receptors, and MC4R is the subtype most often cited as responsible for its behavioural effects. Activity at MC1R, MC3R and MC5R is also reported. Selectivity is therefore described as limited rather than precise.
It is not classified as a vasodilator, and its proposed site of action is the central nervous system rather than peripheral vessels. Some trial participants show small transient blood pressure changes. Whether those changes arise centrally or through another route remains an open question.
Most evidence comes from animal models and from indirect human outcomes such as self-reported desire. Direct measurements of receptor occupancy or signalling in the human brain are not available. Published reviews therefore present a plausible pathway rather than a confirmed sequence of events.
Purity is normally expressed as a percentage of total peak area from a chromatographic run. The value depends on the column, gradient and detection wavelength chosen. Results generated under different conditions are therefore not always interchangeable.
